細胞壁の再生 削除/引用 No.1693-1 - 2009/12/05 (土) 15:58:24 - saya 現在、単細胞の植物をプロトプラスト化し、エレクトロポレーションにて外来遺伝子の導入を試みています。 そこで１つお聞きしたい点があります。 エレクトロポレーション後の細胞に、通常培地を添加すると、 浸透圧の変化によりバーストすると思います。 その為、細胞壁の再生が必要になると思いますが、 その際の浸透圧はどの様に考えれば良いのでしょうか？ つまり、浸透圧を調節しているために添加している、0.8Mマンニトール は存在しても細胞壁は再生するのでしょうか？ それとも、段階的にマンニトール濃度を減少させるなどの操作が必要なのでしょうか？
