I had a trouble about osteoclastogenesis from RAW 264.7 cells for several months ago.
The trouble was no TRAP positive multinucleated cells formation even in "certain" positive control.
At that case, I could troubleshoot by using fresh alpha-MEM, instead old alpha-MEM.
After that, I found the following paper.
Osteoclast Differentiation Requires Ascorbic Acid、J Bone Miner Res 1998;13:970-977
Now I presumed ascorbic acid in old media was depleted and that was the reason of failure.
Such sbsurd problem can not happen in the lab. constantly consumes huge amount of media.
As regards other posi. control, conventional bone marrow culture with 1,25-(OH)2D3 is well-known method.
But I'm not sure if the system suit for Dr. Takao's experiments or not... |
