一つのDH10Bacには1コピーのバクミドしか含まれておらず、ホワイトコロニーであること、目的のタンパク質が産生されていることから、私のコロニーは純粋なコロニーである、ようなことが書かれてあります。
ただ、何度もリストリークするような提案はいただきましたが。取り急ぎ参考までに。
Good to hear you can see the expected band on Western. This for sure confirms the right bacmid was made. However, it is hard to explain why you only get 300bp PCR product.
We tend to think one bacteria cell can only takes one kind of bacmid.This is the basic for cloning, isn't it? If a bacterium can contain both empty vector and ligated plasmid, we won't be able to select the right colonies. This idea should also be applicable to bacmid. Although of course, we don't have any detailed research done on this topic.
Even if one bacterium can contain both bacmids, since the un-transposed bacmid has intact LacZ gene, it should add some Blue color to the cololy. so I don't think this hypothesis can explain everything here.
Usually when we have ambiguious results like this, we recommend to restreak the colonies, because we think this problem is caused by a mixed
population of bacteria, not a single colony. And usually in such cases, after restreak, some blue colonies will show up on the plate. And we will restreak the while colonies again till on a plate we only get white clonies. |
