ones of the U6 and H1 genes, known to synthesize small

in the middle. These shRNAs lack a 5′ cap and a long
polyadenylated tail [31–33]

RNA Pol III promoters, especially
H1 and U6 from human and mouse, have been used
most frequently, since they have a well-defined start site
of transcription and a simple effective termination signal

of small RNA transcripts. Moreover, they can
efficiently transcribe small RNA transcripts lacking both
the 50 cap and 30 polyadenosine [poly(A)] tail [32,33].
