I was able to detect it by extracting with TBS containing 1% TRITON X-100. You are talking about the cyclin inhibitor, right? There are other p21s.
RIPA buffer (TBS containing 0.1 to 1% deoxychalate, 1% TX-100 with or without 0.1%SDS) would also work, but p21 could be released from cyclin complexes. Release of p21 from complexes does not matter for WB analysis. You can also try SDS PAGE sample buffer without dye or 1% SDS in TBS. But, you have to remember that beta-ME and high concentration of glycerol disturb BCA protein assay, and high concentration of SDS is not available for Bradford protein assay. |
