DpnI is able to cut only DNA whose adenine is methylated on the recognition sites. Therefore, DpnI is often used to eliminate or inactivate plasmid DNA after PCR.

My question is that "can DnpI cut hetero-duplex DNA (one of them has methylated adenine)?". During PCR, template DNA may not pair with the original strand.
