You should decide it on case-by-case basis. It depends how big the tissue is, considering the penetration of paraffin. In addition, if you are going to perform immunohistochemistry later, you have to think how stable your target molecule is. As the temperature of paraffin is high, it can affect the antigenity of the molecule.
This way, the shorter, the better, as long as the penetration of paraffin is enough.
If the size of tissue is like 5 mm X 5 mm X 5 mm, 20 min each should be usually enough. |
